TNFα在血管内皮细胞凋亡中的作用

【摘要】目的探讨TNFα引起的血管内皮细胞(VEC)凋亡在皮肤撕脱伤早期血栓形成中的作用. 方法分2组,实验组:内皮细胞(EC)+TNFα,拮抗组:用EC+TNFα+TNFα抗体处理. 采用流式细胞仪分别测定实验组TNFα不同时间、不同浓度以及拮抗组EC凋亡率. 结果 EC凋亡率随TNFα作用时间增加,0~48 h EC凋亡率为(2.6±0.42)%~(40.5±8.3)%;EC凋亡率也随TNFα浓度而增加,0~3000 u*mL-1 EC凋亡率分别为(2.6±0.42)% ~(13.7±2.6)%;TNFα抗体明显拮抗TNFα诱导的EC凋亡(P<0.01). 结论 TNFα可引起EC的凋亡和死亡,具有时间依赖性和浓度依赖性,人重组TNFα单抗可消除因TNFα引起的毒性作用.%AIM To study the effect of TNFα on apoptosis of vascular endothelial cells(VEC) in early second thrombosis after skin avulsion. METHODS It was divided into experimental group (EC+TNFα) and inhibitory group (rhTNFαAb pretreatment of EC+TNFα). According to various times and concentration, apoptosis rate of vascular endothelial cells was measured by FCM for experimental group and inhibitory group. RESULTS TNFα caused massive death of endothelial cells in primary culture in a concentration-and time-dependent manner.
