【摘要】Objective:To screen potential specific biomarkers in serum of gastric cancer patients using functional magnetic bead based sample fractionation
method
combinded
with
matrix-assisted
laser
desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) technology.Methods:The serum samples from 20 gastric cancer patients and 20 healthy volunteers were enriched by the magnetic bead and analyze by MALDI-TOF MS.The results were analyzed by the software FlexAnalysis3.0 and ClinProTools2.1.Results:Seven protein peaks were up regulated in serum of patients with gastric cancer,six protein peaks were down regulated in serum of patients with gastric cancer protein.Two peaks with mass charge ratio (m/z) 2863.71Da and 4965.08Da significantly discriminated gastric cancer from non cancer control group.Conclusion:Using functional magnetic bead based sample fractionation method combined with MALDI-TOF MS can screen potential specific biomarkers in serum of gastric cancer,and it is a usefue method to find and establish potential specific biomarkers for gastric cancer.%目的:应用液体蛋白芯片-飞行时间质谱技术从胃癌患者血清中
筛选潜在的标志蛋白.方法:采用液体蛋白芯片技术和基质辅助激光解吸电离飞行时间质谱技术(MALDI-TOF MS)对20例胃癌患者和20例正常人的血清蛋白谱进行检测,采用FlexAnalysis3.0和ClinProTools2.1软件进行图谱分析和统计学处理.结果:胃癌患者与正常人血清蛋白质谱比较,胃癌患者血清中有7个差异蛋白高表达,6个差异蛋白低表达,其中,差异最显著的两个蛋白的质荷比(m/z)分别是2863.71Da和4965.08Da.结论:利用液体蛋白芯片-飞行时间质谱技术可从血清中筛选出胃癌潜在的标志蛋白,此技术对于发现和筛选血清中的胃癌标志蛋白是一种很有前途的方法. 【总页数】3页(1513-1515)
