【摘要】Objective To investigate the effect of microRNA (miRNA,miR)-181 on osteosar coma cell proliferation and apoptosis and its molecular mechanism.Methods Expression of miR-181 in hmnan osteosarcoma tissues and corresponding normal tissues,normal osteoblast NHOst and osteosarcoma cell lines were detected by Quantitative polymerase chain reaction (qPCR).miR-181 mimics and NC-miRNA were transfected into MG63 or U2OS cells,then CCK-8 assay was used to detect MG63 and U2OS cells proliferation;cell clone formation assay was used to examine the ability of cell cloning;flow cy tometry was performed to determine cell cycles,and the expression of MAP3K10 were detected using Western blotting.The target site of miR-181 was predicted by TargetScan,and was further confirmed by lucif erase activity assay.Further,miR-181 mimics and LV-MAP3K10 were co-transfected into MG63 or U2OS cells,then cells
