金银花精提取物与粗提取物注射液的主动全
身过敏实验研究

【摘要】 目的对不同提取纯化工艺制备的金银花提取物注射液进行免疫毒性实验,以了解含金银花的注射剂致过敏反映的缘故。方式按《中药、天然药物免疫毒性研究的技术指导原那么》的要求,做金银花精提取物与粗提取物注射液的主动全身过敏实验。豚鼠腹腔注射致敏量以绿原酸计别离为, mg·kg-1,静脉注射激发量是致敏量的2倍,激发后详细观看记录每只动物的反映情形。结果金银花精提取物注射液高、低剂量组动物均未显现过敏反映,而金银花粗提取物注射液高剂量组显现类过敏强阳性反映,低剂量组显现阳性反映。结论金银花粗提取物注射液显现类过敏反映的缘故,与注射液中含有蛋白质、树脂、鞣质等杂质有关。

Abstract:ObjectiveTo study the immune toxicity of Honeysuckle extract injection prepared by different extraction and purification processes,in order to analyze the causes of allergic reaction caused by Honeysuckle injection.
MethodsAccording to the requirement of \medicine,natural medicine immunity toxicity study ruller\,we studied active systematic anaphylaxis of Honeysuckle refined extract and crude extract injection. Sensitizing dose(ip) counted as chlorogenic acid was , mg·kg-1, excitation dose(iv) was 2 times of sensitizing dose. The reaction of each animal was observed after excitation. ResultsThe result showed that no allergic reaction occurred in the high and low dose group of Honeysuckle refined extract injection,howerer strong positive reaction ouured in high dose Hoaeysuckle crude injection groups and positive reaction in low dose causes of allergic reaction occurred in Honeysuckle crude extract injection are associated with impurity in the injection such as protein,resin,tannin.

金银花具有清热解毒、抗菌、抗病毒功效。以金银花为主研制的双黄连、清开灵等中药注射剂,被临床普遍用于上呼吸道感染、肺炎等高热疾病。由于疗效靠得住,用量日增,不良反映也就加倍突出。在人们寻觅发生不良反映缘故的时候,有人引用国外文献说金银

1 材料
供试品及其制备方式
金银花粗提取物注射液批号070327,自制。制备方式:金银花2kg(产地:四川古蔺县,经鉴定为忍冬科植物灰毡毛忍冬Lonicera macranthoides Hand.-Mazz.的干燥花蕾)。加水2 000 ml,煎煮3次,1h/次。归并滤液,浓缩至2 000 ml,稍冷,加乙醇使含醇量达75﹪,静置24 h,滤过。醇滤液均分2份,留1份供制备金银花精提取物用,另1份加热挥醇后,加水至1 000 ml,放冷,调pH ,冷藏24 h,滤过,高效液相测绿原酸含量,按每毫升含绿原酸3 mg
