【摘要】[Objective] The study aimed to establish a rapid method to extract the genomic DNA from powdery mildew. [Method] Taking conidia of Erysiphe pisi as materials, genomic DNA was extracted from Erysiphe pisi by Chelex-100. ITS2 was amplification with this extraction as PCR templates. The PCR product of ITS2 gene was cloned and sequenced. [ Result] Chelex-100 extraction method could be an efficient and reliable method for PCR detection. [Conclusion] The research lays the foundation for efficient large-scale molecular classification identification of powdery mildews resources.%[目的]建立快速提取及克隆白粉病菌基因组DNA的方法.[方法]以豌豆白粉菌分生孢子为材料,用Chlex-100法快速提取白粉菌基因组DNA,并以此为模板对内转录间隔区Ⅱ(ITS2)序列进行了巢式PCR扩增、克隆、测序及分析.[结果]Chlex-100法可高效的提取白粉病菌基因组DNA,适用于PCR扩增及相关分析.[结论]为快速有效地对白粉病资源进行大规模分子分类鉴定奠定了基础. 【总页数】2页(22204-22205)
