BET家族基因表达与恶性肿瘤患者总生存期及临床病理特征的关系

[摘要] 目的 探讨新兴肿瘤治疗靶点bromodomain and extra-terminal（BET）家族基因表达与多种实体恶性肿瘤患者预后及临床病理特征的关系。 方法 使用美国Kaplan Meier plotter在线数据库，采集肿瘤样本BET家族基因mRNA水平表达数据及患者临床数据，在线完成生存分析。 结果 胃癌中，BRD2/BRD3/BRDT高表达组患者总生存期显著下降（均P < 0.01）。这一相关性在乳腺癌中仅见于Luminal B型及HER2+型患者。在肺癌中，鳞癌患者生存期与BET家族基因表达无显著相关性，腺癌患者生存与BRD3正相关[风险比95%CI（HR 95%CI）：0.22～0.43]，与BRDT负相关（HR 95%CI：1.08～1.78）。卵巢癌患者预后与BET家族基因表达正相关（BRD2 HR 95%CI：0.70～0.91，BRD3 HR 95%CI：0.76～1.00，BRDT HR 95%CI：0.71～0.93）。 结论 BET家族基因高表达提示胃癌及部分乳腺癌患者预后不良，但在肺癌及卵巢癌中并不提示生存期下降。

[中图分类号] R73 [文献标识码] A [文章编号] 1673-7210（2016）01（b）-0069-05 [Abstract] Objective To evaluate the association between a potential therapeutic target， bromodomain and extra-terminal（BET）family， and the prognosis and clinicopathological features in malignant solid tumor patients. Methods Online cancer database （Kaplan Meier plotter） was used to retrieve data of BET family expression and clinicopathological information， all the survival analysis were performed on the database system. Results Higher expression of BRD2， BRD3 and BRDT were significantly correlated with worse overall survival in gastric patients （all P < 0.01）， while this correlation was only also identified in Luminal B and HER2+ （intrinsic subtype） breast cancer. In lung cancer， squamous cells carcinoma patients' survival were not significantly associated with BET expression， but adenocarcinoma patients' survival were positive correlated with BRD3 [Hazard Ratio 95% CI （HR 95%CI： 0.22-0.43） and negative correlated with BRDT （HR 95%CI： 1.08-1.78）. In ovarian cancer patients， the overall survival was positive correlated with all the three members （BRD2 HR 95%CI： 0.70-0.91，BRD3 HR 95%CI： 0.76-1.00，BRDT HR 95%CI： 0.71-0.93）. Conclusion Higher expression of BET family predicts poor prognosis in gastric cancer patients and HER2+/basal-like breast cancer patients， but not in lung cancer and ovarian cancer patients.
