茶多酚对宰后牦牛肉线粒体细胞凋亡和肌肉嫩度的影响
(1.西南民族大学生命科学与技术学院,成都610041;2.甘肃农业大学食品科学与工程学院,兰州730070)摘要:为探究茶多酚对宰后牦牛肉成熟过程中线粒体氧化应激损伤介导的线粒体细胞凋亡级联反应及对肌肉嫩度的影响,以经茶多酚处理的牦牛背最长肌为研究对象,测定对照组和茶多酚处理组线粒体氧化应激水平、线粒体氧化损伤程度、线粒体功能特性、线粒体细胞凋亡进程,以及肌肉嫩度的变化。结果表明,在宰后成熟早中期,茶多酚组线粒体活性氧(Reactiveoxygenspecies,ROS)水平、丙二醛(Malondialdehyde,MDA)以及羰基含量整体上显著或极显著低于对照组;成熟中后期,线粒体超氧化物歧化酶(Superoxidedismutase,SOD)活性以及线粒体膜流动性显著或极显著高于对照组。24h后,茶多酚组线粒体通透性转换孔(Mitochondrialpermeabilitytransitionpore,MPTP)开放程度显著或极显著低于对照组;6~168h,茶多酚组线粒体Cyt鄄c浓度均高于对照组;72~168h,胞浆细胞色素c(Cytochromec,Cyt鄄c)浓度均显著或极显著高于对照组;在成熟早期,细胞凋亡酶鄄3(Caspase鄄3)活性以及肌原纤维小片化指数(Myofibrillarfragmentationindex,MFI)显著或极显著低于对照组。以上研究结果说明,茶多酚通过抑制ROS介导的氧化应激对线粒体结构和功能的损伤,起到抑制线粒体细胞凋亡级联反应对肌肉嫩度的改善作用,表明茶多酚在发挥抗氧化剂和保鲜剂作用、提高肌肉品质的同时,从细胞凋亡嫩化机制角度出发,对肌肉的嫩化产生不利影响。

Abstract:Aimingtoinvestigatetheeffectsofteapolyphenolsonthemitochondrialapoptosiscascadereactionmediatedbymitochondrialoxidativestressandtendernessofyakmeatduringpostmortemaging.Thelongissimusdorsimusclesinjectedwithinhibitorteapolyphenolweretakenastheexperimentobjects.Thelevelsofmitochondrialoxidativestress,degreeofmitochondrialoxidativedamage,mitochondrialfunctionalcharacteristics,processofmitochondrialapoptosisandchangeofmeattendernessinthecontrolgroupandthetreatmentgroupweremeasured.TheresultsshowedthatthelevelofmitochondrialROS,MDAandcarbonylcontentinthetreatmentgroupwasoverallsignificantlyorextremelysignificantlylowerthanthatinthecontrolgroupintheearlyofpostmortemaging.Atthemiddleandlateofagingtime,theSODactivityandthemitochondrialmembranefluiditywassignificantlyorextremelysignificantlyhigherthanthatinthecontrolgroup.After24h,openingoftheMPTPinthetreatmentgroupwassignificantlyorextremelysignificantlylowerthanthatinthecontrolgroup.After6~168h,theCyt鄄cconcentrationofmitochondriainthetreatmentgroupwashigherthanthatinthecontrolgroup;during72~168h,theCyt鄄cconcentrationofcytoplasminthetreatmentgroupwassignificantlyorextremelysignificantlyhigherthanthatinthecontrolgroup.Meanwhile,Caspase鄄3activityandMFIinthetreatmentgroupwassignificantlyorverysignificantlylowerthanthatinthecontrolgroupatearlyagingtime.Theresearch
