鸡小肠α-葡萄糖苷酶的提取及性质研究

摘 要

α-葡萄糖苷酶广泛存在于自然界的所有生物体内,种类繁多,性质各异。本实验用鸡小肠作原料,经过研磨,搅碎,制成粗酶液,在通过硫酸铵沉淀,透析,大孔树脂层析法提取α-葡萄糖苷酶。以4-硝基苯-α-D-吡喃葡萄糖苷为底物,研究酶催化反应动力学。结果表明,该酶的最适温度范围在30℃~50℃,最适pH范围在6.0~6.5,α-葡萄糖苷酶的热温度性较差。Mg2+、Zn2+、和Fe2+对α-葡萄糖苷酶有激活作用,而Cu2+对α-葡萄糖苷酶有抑制作用。

ABSTRACT

There are various α-glycosidase of different features existing in all kinds of creatures. In this experiment chicken intestinal used as raw material is turn into crude enzymes by grinding, pulperring,and then crude enzymes is changed into precipitation through ammonium sulfate. At last by means of dialysis and macroporous resin chromatography α-glycosidase can be extracted. PNPG can be used to study Enzyme catalysis dynamics. The result suggests that this enzymes is active in temperature 30℃~50℃,pH6.0~6.5 but its hot temperature the gender is poorer. Mg2+、Zn2+、and Fe2+ have activation on α-glycosidase ,while Cu2+ has inhibitory action.
