(广西医科大学,南宁530021)摘要:目的筛选扶芳藤总黄酮的最适提取条件,观察扶芳藤总黄酮提取物对衰老小鼠血清及心肌组织中抗
氧化指标过氧化氢酶(CAT)、总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-Px)活性的影响。方法 通过正 交试验L9(33)法确定扶芳藤总黄酮提取的最适温度、最适料液比以及最适提取时间。取50只健康雄性昆明小鼠,
随机分为正常组、模型组及扶芳藤总黄酮低、中、高剂量组各10只。模型组及扶芳藤总黄酮组给予D-半乳糖0.2 mL/d颈背部皮下注射诱导衰老,正常组注射等量生理盐水,连续1周。从第3周开始,扶芳藤总黄酮低、中、高剂
量组分别给予0.2、0.4、0.8 g/mL扶芳藤总黄酮提取液灌胃,正常对照组给予等量蒸憎水灌胃,连续4周。最后一
次给药后,取小鼠眼球血和心脏,检测血清、心肌组织中的CAT、T-AOC、GSH-Px活性。结果当提取条件为温度 60 C、料液比1:50,时间30 min时,总黄酮提取率为3.57%,高于其他条件下的总黄酮提取率(P均<0.01)。与正
常组比较,模型组血清及心肌组织中CAT、T-AOC、GSH-Px活性下降(PE <0.05);与模型组比较,扶芳藤高、中、低
剂量组血清及心肌组织中CAT、T-AOC、GSH-Px活性均升高(PE <0.05,其中高剂量组各指标高于中、低剂量组 (P均<0. 05) o结论 扶芳藤总黄酮提取的最适条件是:温度60 C、料液比1:50,时间30 mm;扶芳藤总黄酮提取

and to observe the effects of total flavonoids extract of Euonymus fortunei on the catalase ( CAT),total antioxidant capacity
( T-AOC),and glutathione peroxidase ( GSH-Px) activities in the serum and myocardial tissues of aging mice. Methods
The optimum temperature, optimum ratio of material to liquid and optimum extraction time of total flavonoids extracted from
Euonymus fortunei were determined by orthogonal test L9 (33) . Fifty clean Kunming mice were randomly divided into 5
groups: the normal group,model group,low-dose,medium-dose,and high-dose groups of total flavonoid extract of Euony-
mus fortunei,with 10 mice in each group. The mice in the model group and the total flavonoids of Euonymus fortunei
groups were given subcutaneous injection of D-galactose 0. 2 mL/d to induce the aging models, and the mice in the normal group were injected with the same amount of normal saline for 1 week. From the third week on,the low-dose,medium- dose, and high-dose groups of total flavonoids of Euonymus fortunei were given 0. 2, 0. 4, and 0. 8 g/mL total flavonoid ex-tract of Euonymus fortunei,and the normal control group was given the same amount of distilled water for administration,
for 4 consecutive weeks. After the last administration,the CAT,T-AOC and GSH-Px activities in the serum and myocardial
tissues of each group were examined. Results When the extraction temperature was 60 C with extraction ratio of 1: 50
