农杆菌介导棉花茎尖遗传转化体系优化

【摘要】生根困难和转化效率较低是农杆菌介导棉花茎尖遗传转化体系的应用瓶颈.本研究以柯字棉312为研究材料,探讨茎尖遗传转化中植物生长调节剂、生根诱导时间、无菌苗型、针刺处理等对茎尖生根率、茎尖形态以及抗性苗产生率等的影响.结果表明,1.5 mg/L NAA能够有效提高生根率,促进茎尖伸长;生根诱导时间是生根率的重要影响因子;经多次针刺处理的正常苗抗性苗率较高;培养5 d的正常苗茎尖生长速度和状态优于黄化苗.%Low root-inducing rate and transformation efficiency are the tough problems affecting Agrobacterium-mediated genetic transformation of cotton shoot apex system.Using shoot apex of Coker 312,this paper investigated the effect of plant growth regulators, root-inducing time, different kind of sterile seedlings and acupuncture on the root-inducing rate, transformation on efficiency and morphology.The result showed that 1.5 mg/L NAA could not only promote root-inducing rate but also accelerate its growing speed; Foot-inducing time was an important factor affecting root-inducing rate.In addition,acupuncture shoot apex could increase the occuring rate of Kan-risitant plant, and the transformation efficiency of the the shoot apex from 5 day sterile seedlings was higher than that from the etiolated ones.
