根霉与米曲霉原生质体一次基因组改组后代酶
活力和形态特性变化规律研究

摘 要
本实验是对根霉与米曲霉原生质体一次基因改组的后代进行进一步的研究,经人工培养多代后观察菌株的形态特征、部分生理生化特性分析。实验对亲本米曲霉、根霉、及其融合后的后代进行了传代培养。通过对亲本以及一、三、五、七代在生长时的形态特征进行定量分析,结果表明亲本和其杂交后代在培养基上生长都比较旺盛,菌体形状颜色都比较相似。测定蛋白酶、淀粉霉的活力,淀粉霉活力中亲本根霉的平均Hc值是1.527,亲本米曲霉的平均Hc值是1.518,而后代霉活最高的融合一代3的平均Hc值是1.781,最低的融合七代1的平均Hc值是1.193。SDS电泳分析发现后代的条带大多比亲本少,说明后代与亲本存在一定差异。

ABSTRACT
This experiment is Rhizopus oryzae protoplasts with the restructuring of the descendants of a gene for further study, after several generations of artificial culture strains were observed morphological, physiological and biochemical characteristics analysis.Experiments on the parent Aspergillus,Rhizopus, and their descendants were fused subculture.By parents,and one, three, five, seven generations in the growth morphology when quantitative analysis shows that parents and their hybrid offspring are more vigorous growth medium, the cell shape of color are more similar. Determination of protease, amylase, starch mold activity in the parental Rhizopus average Hc value is 1.527, the parent Aspergillus average Hc value is 1.518, while the descendants of mold living the highest integration of generation 3, the average Hc value is 1.781, the lowest fusion seven generations of an average Hc value is 1.193. Finally its SDS gel electrophoresis analysis showed that most bands of future generations less than the parents, offspring and parents that there are some differences.
