HPLC法测定导赤丸中黄芩苷和大黄素的含量

【摘要】目的:建立测定导赤丸中2种有效成分黄芩苷和大黄素的含量的方法.方法:采用高效液相色谱法.色谱柱为Chromatrex C18柱;测定黄芩苷的流动相为甲醇-水(加入磷酸调pH=3.5,43:57),检测波长为280nm,流速为1.0mL·min-1,柱温为30℃;测定大黄素的流动相为甲醇-水(加入冰醋酸调pH=2.0,80:20),检测波长为245nm,流速为1.0 mL·min-1,柱温为25℃.结果:黄芩苷和大黄素的检测浓度分别在0.626~1.044mg·mL-1(r=0.9996)和5~25μg·mL-1(r=0.9998)范围内线性关系良好;平均回收率(n=9)分别为100.18%(RSD=0.75%)和99.30%(RSD=1.5%).结论:本方法操作简便、准确度高、专属性强,适用于导赤丸的质量控制.%OBJECTIVE: To establish the method for the content determination of baicalin and emodin in Daochi pills. METHODS: HPLC was adopted with chromatrex C18 as column. The mobile phase for baicalin determination consisted of methanol-water-phosphoric acid (43∶57) with detection wavelength at 280 nm. The flow rate was set at 1.0 mL·min-1 and the column temperature was 30 ℃ .The mobile phase for emodin determination was methanol-water-glacial acetic acid (80∶20) with detection wavelength at 245 nm.
