丁苯酞对脑低灌注老龄大鼠胆碱能神经功能的影响

【摘要】Objective To observe the improving effects of butylphthalide (NBP) on the learning and memory of cerebral hypoper-fused rats and investigate its effects on choline acetyltransferase( ChAT) and acetylcholine esterase( AChE) activity. Methods Cerebral hy-poperfused models were made with ligating rats' bilateral common carotid arteries for three months. There were four groups;sham, vehicle and NBP 30 and 120 mg/kg. The last two groups were administered NBP for 45 days consecutively. Then rats' spatial learning and memory were measured with Morris water maze and the ChAT and AChE activity in rats' brains were measured with biochemical method. Results In water maze positioning navigation trial, NBP reduced significantly the latencies of cerebral hypoperfused rats finding safe platform. In spatial exploring trial, NBP increased cerebral hypoperfused rats' time percentages spent in the platform-quadrant Meanwhile, NBP increased the ChAT activity in cerebral hypoperfused rats'cortex and hippocampus, but couldn't impact rats' brain AChE activity. Conclusions NBP could improve cerebral hypoperfused rats' learning and memory deficits. Increasing the ChAT activity and amelioratingcholinergic system in brain is one of
