【摘要】Objective To study the effects of parathyroid hormone ( PTH) combined with zoledronic acid (ZA) on fracture healing in ovariectomized mice. Methods Ninety six 12-week C57BL/6 female mice were enrolled in this study. These mice received transverse fracture at the lower third of left femur after being ovariectomized one month lat-er. The mice were then randomly divided into four groups and treated with normal saline, PTH1-34, zoledronic acid (ZA), and PTH1-34 plus ZA. Eight mice were killed at the second and fourth week respectively in each group. Micro-CT, biomechanical test, histology and RT-PCR were performed. Results At the 2nd week after administration, bone mineral density (BMD) [ (0. 062±0. 009) g/cm3], bone volume (BV) [ (34. 8±2. 6) mm3], relative bone volume (BV/TV)[ (52. 3±3. 0)%] and trabecular bone thickness (Tb. Th) [ (0. 212±0. 031) μm] in PTH1-34 plus ZA group were significantly higher than saline group (P<0. 05). At the 4th week after administration, BMD[ (0. 072± 0. 010) g/cm3] and BV/TV[ (60. 3±5. 2)%] were significantly higher than other groups (P<0. 05).
