【Abstract】 AIM: To investigate the effects of adult bone marrow mesenchymal stem cells on the expansion of umbilical cord blood CD34+ cells ex vivo. METHODS: Adult bone marrow mesenchymal stem cells were isolated and cultured as feeder layer. Umbilical cord blood CD34+ cells were cultured in combination with mesenchymal stem cells, stem cell factor, Interleukin11 and granulocytemacrophage colony stimulating factor, respectively. At the end of 3 weeks, the total number of nucleated cells , colony forming cells and CD34+ cells was repeatedly counted in culture system for 3 weeks to obtain the fold of expansion. RESULTS: The mesenchymal stem cells and cytokines coculture systems significantly enhanced the expansion of the cord blood cells with the increase of the total number of nucleated cells by ±, colony forming cells by ± and CD34+ cells by ±, respectively. CONCLUSION: The expansion of umbilical cord blood CD34+ cells can be enhanced ex vivo by adult
bone marrow mesenchymal stem cells combined with cytokines.

【摘要】 目的: 探讨骨髓间充质干细胞(MSCS)对脐血CD34+细胞体外扩增作用. 方式: 分离培育成人MSCS作为滋养层,联合SCF, IL11和GMCSF别离组成对脐血CD34+细胞的不同扩增体系,培育3 wk后观看脐血有核细胞、CFCs和CD34+细胞扩增倍数的转变. 结果: MSCS联合细胞因子组较其他组培育体系对有核细胞总数、CFCs含量及CD34+细胞含量均具有明显的扩增作用,别离扩增了±, ±, ± 倍. 结论: 成人MSCS协同其他细胞因子可增强脐血CD34+细胞的体外扩增作用.

0引言
骨髓间充质干细胞(mesenchymal stem cells, MSCs)作为骨髓造血微环境中的一种重要细胞成份,最近几年来成为干细胞研究领域的一大热点. MSCs具有自我更新和多向分化潜能,在体外可被诱导分化为多种结缔组织及部份来源于外胚层的组织,形成骨、软骨、骨骼肌、腱、韧带、真皮、脂肪、骨髓基质和神经[1,2]. 另外,还分泌多种生长因子支持骨髓造血,藉此MSCs可在体外作为滋养细胞层进行造血干/祖细胞扩增. 自脐血(umbilical cord blood,UCB)中发觉造血干细胞(hematopoietic stem cell,HSC)以来,脐血HSC移植用
于医治恶性及非恶性血液病日趋普遍,但单份脐血所含的造血细胞不足以知足成人的移植所需,大量扩增后才能用于移植. 鉴于此,咱们探讨了来自成人的MSCS对脐血CD34+ 细胞的体外扩增作用.
1材料和方式 材料
正常成年男性献骨髓者2例,健康足月临盆儿脐血14例,取自兰州市慈和堂医院和本院妇产科. Percoll分离液(密度 kg/L),淋巴细胞分离液(密度 kg/L),马血清(HS)均购自TBD 公司;DMEMLG培育基购自PAA Lab (Germany);IMDM购自Gibco公司;胎牛血清(FBS)为杭州四季青公司产品;BSA、甲基纤维素、β巯基乙醇及L谷氨酰胺购自Sigma公司;重组人干细胞因子(rhSCF),重组人白细胞介素11(rhIL11), 重组人粒细胞巨噬细胞集落刺激因子(rhGMCSF),重组人粒细胞集落刺激因子(rhGCSF),重组人红细胞生成素(rhEPO)均购自Peprotech公司;荧光标记的抗CD34单克隆抗体(CD34PE)购自Coulter Immunotech公司;抗CD29, CD44, CD45和抗HLADR单克隆抗体购自PharMingen公司.
方式
的分离纯化及培育MSCs的分离纯化及培育按文献[3]介绍的方式进行. 搜集正常肝素抗凝骨髓约10 mL,用Percoll溶液梯度离心,900 g 30 min,搜集中间相为有核细胞,然后将细胞按2×108 /L密度接种于DMEMLG培育基 (含100 mL/L FBS, L谷氨酰胺,青霉素105 u/L,庆大霉素8×104 u/L)的塑料培育瓶中,37℃, 50 mL/L CO2
