【摘要】Aim:To investigate protective effect of recombinant Elafin on CD 14 receptor of macrophage challenged by polymorphonuclear granulocyte ( PMN) and lipopolysaccharide ( LPS). Methods: Plasmid pEGFP-C1-Elafin was transfected into macrophage, then challenged for 4h by PMN and UPS. Expressed Elafin was detected by inverted fluorescence microscope and CD14 was determined by flow cytometry. Transfected macrophage cocultured for 1h with apoptotic PMN to observe the number of macrophage interacted with apoptotic PMN. Results: Transfected pEGFP-C1-Elafin was expressed successfully in macro-phage , of which the CD14 quantity increased in evidence. And the amount of macrophage combined with apoptic PMN was also elevated. Conclusion: The approach of genetic modification of macrophage with Elafin can be used to restore apoptotic related receptor CD14 in inflammatory condition.%目的:研究转染重组Elafin对炎症状态下巨噬细胞表面受体CD14的保护作用.方法:重组质粒pEG-FP-C1-Elafin转染入离体培养巨噬细胞株,与多形核中性粒细胞(polymorphonuclear granulocyte,PMN)、脂多糖(lipopolysaccharide,LPS)共作用4h后,倒置荧光
