四臂星型聚乙二醇/聚乳酸的合成及其负载羟基喜树碱研

【摘要】4-armed star poly ( ethylene glycol )/polylactide copolymers ( 4SPEG/PLA ) were synthesized by ring-opening polymerization of lactide ( LA ) with 4-armed poly ( ethylene glycol ) as macromolecular initiator and stannous octoate as catalyst.Hydroxycamptothecin ( HCPT )-loaded nanoparticles made from 4SPEG/PLA were prepared by dialysis method.The size and size distribution, drug loading content, stability, in vitro drug release and in vitro cell cytotoxicity of HCPT-loaded nanoparticles were studied.It was found that the size distribution was narrow and the average diameter was below 200 nm.In vitro drug release experiments indicated that HCPT-loaded nanoparticles showed sustained release profile.Cytotocicity tests showed that the cytotoxicity of HCPT-loaded nanoparticles against cancer cell in vitro was remarkably higher than that of free drugs.These data demonstrated that such HCPT-loaded nanoparticles may serve as a stable delivery system for poorly soluble HCPT.%以四臂聚乙二醇为引发剂,辛酸亚锡为催化剂,通过开环聚合反应制备四臂星型聚乙二醇/聚乳酸共聚物。通过透析法制备了负载抗肿瘤药物羟基喜树碱的四臂星型聚乙二醇/聚乳酸纳米粒子,并对其粒径
