HPLC-ELSD法测定散结通胶囊中胆酸的含量

【摘要】目的 制定高效液相色谱-蒸发光散射检测法测定散结通胶囊中胆酸的含量.方法 采用Kromasil 100-5 C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇-0.1%冰醋酸(75:25)为流动相,流速1.0 mL/min,进样体积20μL;蒸发光散射检测器参数:漂移管温度40℃,载气(N2)压力200 kPa.结果 胆酸进样量在1.368~13.68μg范围内,进样量的对数值与峰面积的对数值呈良好的线性关系(r=0.9999),平均加样回收率为100.26%(RSD=2.4%,n=6).结论 本方法简便准确,可用于散结通胶囊的质量控制.%Objective To establish an HPLC-ELSD method for determination of cholic acid in Sanjietong Capsules. Methods HPLC was performed on a Kromasil 100-5 C18 column (250 mm×4.6 mm, 5 μm), and the mobile phase was methanol-0.1 % glacial acetic (75:25) at a flow rate of 1.0 mL/min. The sample volume was 20 μL. The parameters of ELSD were set as follows: the evaporation temperature was 40 ℃, and the carrier gas (N2) pressure was 200 kpa. Results A good linear correlation of logarithm of sample quantity with peak area was observed within the range of 1.368-13.680 μg for cholic acid (r=0.9999). The average recovery was 100.26 %, with RSD of 2.4 % (n=6). Conclusion The method is convenient and accurate, and can be used for the quality control of Sanjietong Capsules.
